Charaka Samhita, Chikitsa Sthana 1.1.7-8, describes benefits traditionally attributed to properly used Rasayana, including longevity, memory, intellect, health, youthfulness, complexion, voice, and strength, and defines it as a means of obtaining excellent quality of rasa and the other dhatus. This is a classical therapeutic framework; it should not be retrospectively translated as “telomerase activation,” “DNA repair,” or “epigenetic rejuvenation” unless those mechanisms are demonstrated experimentally.
Ashwagandha: Direct Telomere Evidence Is In Vitro
The strongest direct telomerase-related Ashwagandha report found is a 2016 laboratory study, not a human trial. Withania somnifera root extract was applied for 72 hours to HeLa cervical-cancer cells, and TRAP-assay telomerase activity rose by about 45% at selected concentrations. Because HeLa cells already possess active telomerase and differ from normal adult tissues, this cannot show that oral Ashwagandha lengthens human telomeres, slows ageing, or extends life.
Human research does provide a different, narrower signal. Randomized trials and NIH reviews indicate that some Ashwagandha extracts may reduce perceived stress and cortisol and may improve sleep over short periods. Because stress and telomere biology can be associated observationally, stress reduction is a plausible research hypothesis, not proof of telomere preservation. The Ayurvedic Pharmacopoeia of India identifies the root of Withania somnifera and lists its properties as tikta-kashaya rasa, laghu guna, ushna virya, madhura vipaka, with Rasayana, balya, vajikarana, and Vata-Kapha-pacifying actions. These classical descriptors should remain distinct from modern molecular endpoints.
Amalaki: Classical Rasayana, No Verified Epigenetic Clock Trial
Amalaki or Amla is firmly documented as a Rasayana drug in the Ayurvedic Pharmacopoeia of India. For the fruit, the Pharmacopoeia lists five tastes—madhura, amla, katu, tikta, and kashaya—together with laghu and ruksha guna, shita virya, madhura vipaka, and actions including Rasayana and tridoshajit. It also names Cyavanaprasha as an important formulation. This supports Amalaki’s classical rejuvenative status without requiring a modern telomere claim.
Human trials of Phyllanthus emblica extracts have examined outcomes such as lipids, endothelial function, oxidative-stress markers, inflammation, and glucose-related measures. Vitamin C content differs with cultivar, maturity, storage, processing, and assay. The Pharmacopoeia confirms ascorbic acid and tannins among Amalaki constituents but does not endorse a fixed “20-30 times orange” comparison.
Brahmi and Bacopa: Cognitive Research Is Not Telomere Research
Bacopa monnieri is widely marketed as Brahmi and has been studied in randomized trials for memory and cognitive performance, including a placebo-controlled study in healthy adults aged 65 or older. The available human Bacopa literature does not demonstrate telomere lengthening, senescent-cell clearance, or lifespan extension.
Classical naming also requires care. Both Bacopa monnieri and Centella asiatica have been called Brahmi in different regions and commercial contexts. In discussions of Charaka’s four Medhya Rasayana drugs, Mandukaparni is generally identified as Centella asiatica, while Guduchi is Tinospora cordifolia. It is therefore inaccurate to use a modern Bacopa extract trial as automatic confirmation of every classical reference to Brahmi or Mandukaparni.
Guduchi: Traditional Use Does Not Establish “Immunosenescence Reversal”
Guduchi is included in classical Medhya Rasayana discussions. Laboratory and animal studies describe immunomodulatory and anti-inflammatory effects, yet authoritative clinical reviews note that such activities have not been established as anti-ageing benefits in humans. There is no credible evidence that Guduchi reverses immune ageing or preserves lymphocyte telomeres.
Safety is especially important. Published case series and the NIH LiverTox monograph associate Tinospora cordifolia products with clinically apparent liver injury, often with autoimmune-like features, and risk may be greater in people with pre-existing liver disease. This does not erase its classical use, but it makes unsupervised long-term “immune boosting” or anti-ageing dosing inappropriate.

Classical Ayurvedic physicians did not describe chromosomes, telomerase, DNA methylation clocks, FOXO3a, SIRT1, or mTOR. They described observed qualities, indications, preparation methods, eligibility, diet, conduct, and supervised use within an Ayurvedic model of dosha, agni, srotas, dhatu, and ojas. Without these steps, molecular language may make traditional claims sound more proven than they are.
